To distinguish the two meanings, pay attention to the verbs and the context. If it is about membership and shared interests ('beitreten' (to join), 'Sport' (sports)), it means an association or club. If it is about going out and music ('tanzen' (to dance), 'feiern' (to party)), it means a nightclub.

When 'Klub' means an association or club, it is often used with verbs such as 'einem Klub beitreten' (to join a club), 'einen Klub gründen' (to found a club), or 'Mitglied in einem Klub sein' (to be a member of a club). The type of club is also often named, e.g. 'Sportklub' (sports club), 'Buchklub' (book club).

When a nightclub is meant, the sentence usually describes movement there or being there. Typical formulations are 'in den Klub gehen' (to go to the club), 'in einem Klub tanzen' (to dance in a club), or 'die ganze Nacht im Klub feiern' (to party all night at the club).

club / association
Verein and Klub can both refer to an organized group with shared interests. Verein tends to emphasize a formal member organization, whereas Klub can place more emphasis on like-minded people meeting regularly. With sports and leisure groups, however, the traditional name often determines which word is used.
