'die Lage' and 'die Konstellation' can both describe the circumstances in a situation. 'die Lage' is more general and fits very often when talking about the current situation, for example in a family, at work, or in a crisis. 'die Konstellation' puts more emphasis on how different factors or people interact. The word sounds somewhat more precise and often more formal.
