The English form of address for a man, corresponding to the German 'Herr' (Mr.). In German, it is mostly used when talking about English-speaking people or in an international context.
Der Chef der Delegation ist ein gewisser Mister Smith aus London.
The head of the delegation is a certain Mister Smith from London.
You can recognize the meaning from the context: If 'Mister' comes before a surname (e.g. 'Mister Jones'), it is the form of address. If it comes before a geographical name (e.g. 'Mister Germany'), it is a competition title.

The new Mister Austria was crowned yesterday evening.
As a form of address, 'Mister' in German is used almost only for men from the English-speaking world. It is unusual to address a German person in everyday life as 'Mister Schmidt', except in a very international or ironic context.

'der Herr' and 'der Mister' can both be understood as forms of address for a man. 'Herr' is the German form and is used completely normally in German with the last name. 'Mister' is the English form of address and is used in German mainly when talking about English-speaking people or when an international context is meant. Learners may confuse the words because 'Mister' seems like the English equivalent of 'Herr'. But in a normal German conversation, you say 'Herr Becker', not 'Mister Becker'.
